灵芝多糖真空耦合超声提取工艺优化及其抗帕金森病活性研究

摘要: 为了提高灵芝多糖的提取率,并评价其抗帕金森病(Parkinson's disease, PD)活性,文章通过单因素实验和正交试验优化灵芝多糖的真空耦合超声提取工艺,并采用1-甲基-4-苯基-1,2,3,6-四氢吡啶(MPTP)诱导的PD小鼠模型评价灵芝多糖的抗PD活性。结果表明:灵芝多糖真空耦合超声提取的最佳工艺条件为超声时间120 min、超声温度50 ℃、超声功率300 W、料液比1:30;在该条件下,灵芝多糖提取率为2.16%,与超声辅助提取法相比,灵芝多糖提取率提高了52.11%。体内抗PD活性实验结果显示,灵芝多糖可显著改善PD小鼠的运动功能障碍,并可抑制多巴胺能神经元变性死亡。研究结果表明,真空耦合超声提取法能显著提高灵芝多糖的提取率,且灵芝多糖具有显著的抗PD活性。

Abstract: To enhance the extraction yield of Ganoderma lucidum polysaccharides, and to evaluate their anti-Parkinson's disease(PD) activity, vacuum-coupled ultrasonic extraction conditions of G. lucidum polysaccharides were optimized by single factor test and orthogonal test, and the anti-PD activity of G. lucidum polysaccharides was evaluated in 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP) induced PD mouse model. The optimal extraction conditions were as follows: ultrasonic time of 120 min, ultrasonic temperature of 50 $ {}^{\circ} $C, ultrasonic power of 300 W, and solid-liquid ratio of 1:30. Under these conditions, the extraction yield of G. lucidum polysaccharides was 2.16%. Compared with ultrasound-assisted extraction, the extraction yield of G. lucidum polysaccharides was enhanced by 52.11% via vacuum-coupled ultrasonic extraction. The in vivo anti-PD activity evaluations demonstrated that G. lucidum polysaccharides could significantly improve the motor dysfunction and inhibit the dopaminergic neuron loss of PD mice. These results prove that vacuum-coupled ultrasonic extraction can be used to enhance the extraction yield of G. lucidum polysaccharides, and G. lucidum polysaccharides exhibit significant anti-PD activity.
