Abstract
To enhance the extraction yield of Ganoderma lucidum polysaccharides, and to evaluate their anti-Parkinson's disease(PD) activity, vacuum-coupled ultrasonic extraction conditions of G. lucidum polysaccharides were optimized by single factor test and orthogonal test, and the anti-PD activity of G. lucidum polysaccharides was evaluated in 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP) induced PD mouse model. The optimal extraction conditions were as follows: ultrasonic time of 120 min, ultrasonic temperature of 50 $ {}^{\circ} $C, ultrasonic power of 300 W, and solid-liquid ratio of 1:30. Under these conditions, the extraction yield of G. lucidum polysaccharides was 2.16%. Compared with ultrasound-assisted extraction, the extraction yield of G. lucidum polysaccharides was enhanced by 52.11% via vacuum-coupled ultrasonic extraction. The in vivo anti-PD activity evaluations demonstrated that G. lucidum polysaccharides could significantly improve the motor dysfunction and inhibit the dopaminergic neuron loss of PD mice. These results prove that vacuum-coupled ultrasonic extraction can be used to enhance the extraction yield of G. lucidum polysaccharides, and G. lucidum polysaccharides exhibit significant anti-PD activity.
