SlERFD3 在番茄果实成熟过程中的功能研究

摘要
为了探究 SlERFD3 在番茄果实成熟过程中发挥的作用,文章分析了 SlERFD3 在番茄果实成熟不同时期表达量的变化情况,利用病毒诱导基因沉默技术(virus-induced gene silencing,VIGS)构建 SlERFD3 沉默番茄植株,发现 SlERFD3 沉默加快了番茄成熟进程;检测了 SlERFD3 沉默番茄果实中叶绿素、类胡萝卜素的质量比,结果发现,与对照组相比,SlERFD3 沉默使番茄叶绿素质量比减少、类胡萝卜素质量比增多;利用实时荧光定量聚合酶链式反应(polymerase chain reaction,PCR)技术检测 SlERFD3 沉默番茄中叶绿素降解基因 NYC1、PAO 的表达量和类胡萝卜素合成基因 PSY1 的表达量,结果发现,与对照组相比,基因 NYC1、PAO、PSY1 表达量均升高,表明 SlERFD3 在番茄成熟过程中作为负调控因子发挥作用。

Functional study of SlERFD3 in tomato fruit ripening

Abstract
To investigate the role played by SlERFD3 in tomato fruit ripening, this paper first analyzed the changes in SlERFD3 expression at different periods of tomato fruit ripening. SlERFD3-silenced tomato plants were constructed using virus-induced gene silencing (VIGS), and it was found that SlERFD3 silencing accelerated the tomato ripening process. Thereafter, the chlorophyll and carotenoid mass ratios of SlERFD3-silenced tomato fruits were examined, and it was found that SlERFD3 silencing reduced the chlorophyll mass ratios and increased the carotenoid mass ratios of tomatoes compared to the control group. Further detection of chlorophyll degradation genes NYC1 and PAO and carotenoid synthesis gene PSY1 in SlERFD3-silenced tomatoes using real-time quantitative polymerase chain reaction (PCR) technique revealed that the expressions of genes NYC1, PAO and PSY1 were higher than those in the control group, indicating that SlERFD3 acts as a negative regulator in the ripening process of tomato.
