基于 UPLC-Q-Orbitrap HRMS 的痂状炭角菌子实体化学成分分析

摘要
痈状炭角菌(Xylaria sp. L1)是一种高价值药用真菌,属于炭角菌科,它生长在野外废弃的白蚁巢穴周围。文章应用超高效液相色谱(ultra-high performance liquid chromatography, UPLC)-四极杆-静电场轨道阱高分辨质谱(quadrupole-orbitrap high-resolution mass spectrometry, Q-Orbitrap HRMS)技术对痈状炭角菌子实体80%甲醇提取物的化学成分进行定性分析。采用Welch RP-C $ _{18} $柱色谱(150 mm×2.1 mm, 1.8 μm),以0.1%甲酸水溶液和0.1%甲酸乙腈为流动相进行梯度洗脱;高分辨质谱分析采用电喷雾离子源(electrospray ionization source, ESI),在正、负离子模式下扫描采集数据;从痈状炭角菌子实体中共检测到41种化合物,包含6种萜类化合物、4种苯丙素类化合物、14种生物碱类化合物、10种长链不饱和脂肪酸类化合物、7种甾体类化合物,其中有18种化合物为痈状炭角菌子实体中首次发现。该研究对痈状炭角菌子实体化学成分进行较全面地分析,为该真菌的深入研究奠定基础。

Analysis and identification of chemical components of Xylaria sp. L1 sporocarps by UPLC-Q-Orbitrap HRMS

Abstract
Xylaria sp. L1 is a high value medicinal fungus belonging to the family of Xylariaceae. It is found growing in wilds around the abandoned termite nests. In this experiment, the chemical components of Xylaria sp. L1 sporocarps extracted with 80% methanol were analyzed and identified by ultra-high performance liquid chromatography coupled with hybrid quadrupole-orbitrap high-resolution mass spectrometry (UPLC-Q-Orbitrap HRMS) method. Compounds were performed on Welch RP-C $ _{18} $ column (150 mm×2.1 mm, 1.8 μm) with a gradient elution of acetonitrile-water containing 0.1% formic acid. The data were collected in positive and negative ion modes using electron spray with electrospray ionization source (ESI). A total of 41 components were identified including six terpenoids, four phenylpropanoids, fourteen alkaloids, ten long chain unsaturated fatty acids and seven steroids components. Among them, 18 compounds were firstly identified in Xylaria sp. L1 sporocarps. The study has quite completely identified the chemical components in Xylaria sp. L1 sporocarps, which may lay the foundation for further study and utilization of the fungus.
